Some of my favorite movies involving a mad scientist character are Bride of the Monster (1955), The Fly (1958 or 1986), Re-Animator (1985) and Frankenhooker (1990).

Be warned, Bride of the Monster is an Ed Wood film, so it's not great technically speaking, but it has a ton of charm!

Peter Lorre was a doctor and definitely mad, in "Mad Love".

I also like Karloff's mad doc movies for Columbia.

The Man They Could Not Hang (1939)
The Man with Nine Lives (1940)
Before I Hang (1940)
The Devil Commands (1941)

He's not "Classic" but one of my favorite Mad Doctors is Dr Emilio Lizardo from BUCKAROO BANZAI, portrayed by the great John Lithgow.

And of course DR STRANGELOVE, even though he isn't in the film for very long.

I can' think mad doctor without including Herbert West from Re-Animator. A modern classic. Be warned though, this movie is not for the squeamish.
